Abaloparatide is a PTHrP(1-34) analog acting as an agonist at the PTH1 receptor (PTH1R). It activates the cAMP signaling pathway to stimulate osteoblastic activity and new bone formation. Administered as 80 mcg subcutaneously once daily. Abaloparatide demonstrates rapid and robust bone formation with the bone turnover marker sP1NP peaking at 93% increase by Month 1, and achieves an 86% relative risk reduction in new vertebral fractures at 18 months in postmenopausal women with osteoporosis at high risk for fracture. Additionally approved for increasing bone mineral density in men with osteoporosis, abaloparatide offers a potent anabolic treatment option that fundamentally enhances bone architecture through PTH1 receptor-mediated osteoblastic activation.
