Orforglipron is an orally bioavailable, non-peptide glucagon-like peptide-1 receptor (GLP-1R) agonist. It has demonstrated potent agonist activity at the human GLP-1R, with an EC₅₀ in the low nanomolar range (~0.5–1 nM) in cAMP accumulation assays in HEK293 cells expressing GLP-1R. In clinical studies, Orforglipron significantly reduced body weight and fasting plasma glucose levels in a dose-dependent manner. Additionally, it showed favorable pharmacokinetics, with a half-life supportive of once-daily dosing and no need for injection, distinguishing it from peptide-based GLP-1 agonists.
